      Crizalk 250mg only acts entirely in cancer cells which have an overactive version of ALK. In 100 people (5%) About 5 with NSCLC have this. It is known as ALK positive disease

INDICATION

     Crizalk 250mg is an antineoplastic drug which is;
Indicated for treatment in patients who suffering from locally advanced or metastatic non-small cell lung cancer (NSCLC) which is anaplastic lymphoma kinase-positive as detected by an FDA-approved test
Indicated for metastatic NSCLC tumors that are ROS1-positive.

MECHANISM OF ACTION

       Prohibitor of receptor tyrosine kinases containing ALK, Hepatocyte Growth Factor Receptor (HGFR, c-Met), and Recepteur d’Origine Nantais (RON)
The gene’s expression and signaling which contribute to raised cell multiplication and continuation of the tumors become stimulate following the expression of ALK oncogenic fusion proteins
Prohibits the signaling whichhelp the expression of these oncogenic fusion proteins, hencepreventing tumor cell proliferation

DOSAGE MANAGEMENT

       Some patients are selected for the treatment with Crizalk based on ALK or ROS1 presences in tumor specimens
The recommended dosage of Crizalk is 250mg orally twice in a day
Crizalk can be taken with or without food, until disease progression
The capsule Crizalk should be swallowed whole.
Crizalk should not break, open, or chew the capsule.
Dosage modification :
Crizalk 200 mg taken orally twice daily for First dose reduction
Crizalk 250 mg taken orally once daily for Second dose reduction
Permanently discontinue if unable to tolerate Crizalk 250 mg taken orally once daily.

PRECAUTIONS

     When administrating Crizalk 250mg,Hepatotoxicity resulted, increase in ALT and total bilirubin resulted, regulate q2wk for 2 months, then monthly
Using Crizalk 250mg capsules will causes Interstitial lung disease/pneumonitis.
When Crizalk 250mg administratingQT interval prolongation resulted, do not use congenital long QT syndrome.
While on Crizalk 250mg treatment, Symptomatic bradycardia resulted, containing syncope; do not use Crizalk 250mg combination with other drugs known to cause bradycardia; regulate heart rate and blood pressure regularly.
Serious visual loss is reported.
While administrating Crizalk 250mg to pregnant womenEmbryo-fetal toxicity will occur.

PREGNANCY

      Pregnancy category D: Crizalk 250mg will cause fetal harm when given to pregnant women
Advice the female not to become pregnant while on treatment with Crizalk 250mg

LACTATION

While therapy with Crizalk 250mg, advise the women not to breastfed to infants
